ANTIFUNGAL DRUG RESISTANT FUNGI ASSOCIATED WITH FARM SOIL IN MBIRI, DELTA STATE

ABSTRACT/DESCRIPTION


Fungal diseases constitute a substantial threat to several plant and animal species as well as to public health. Farm soils have been the highest reservoir of these fungal pathogens. Antifungal drugs treat fungal infections by killing or stopping the growth of dangerous fungi in the body. Fungi, like bacteria, can develop antibiotic resistance, when germs like bacteria and fungi develop the ability to defeat the drugs designed to kill them. Antifungal resistance occurs when fungi no longer respond to antifungal drugs. In order to ascertain antifungal drug resistance to fungi in farm soils in Mbiri, Delta state, this study was carried out. A total of 15 soil samples, collected from top 0cm to bottom 20cm with a sterile spatula, were collected from 3 points in 5 farm soils in the study area. These samples were labelled accordingly and transported in a leak-proof bag to the laboratory for microbial analysis. Two (2)g of each soil sample were weighed and put into a 15ml falcon tube. 8ml of 0.85% of NaCl and 4ml of 0.01% of Tween 20 were dispensed into the falcon tube and the solution vigorously vortexed and allowed to settle for 1 minute. Using a micropipette, 100 microlitre of the supernatant was inoculated onto Sabouraud Dextrose Agar (SDA) with Ketoconazole (16mg/L) and SDA without the drug (in duplicates) using the spread plate method. The total of 45 inoculated plates were incubated at 30ÂșC for 3 - 7 days. The percentage of resistance species was observed and percentage resistance calculated. The mold isolates were purified by subculturing onto SDA plates. The isolates were identified macroscopically and microscopically. The percentage frequency of occurrence was recorded. 70 isolates including 5 different species of Aspergillus were identified. 60% of the species showed resistance to ketoconazole. The percentage frequency of occurrence showed that one sp of Aspergillus occurred the most with 34.3% while another Aspergillus sp occured the least with 12.9%. This study shows there are resistant fungi in farm soils in my study area which can pose threat to crops and humans. Therefore, it is advisable that more studies should be carried out on these fields to help know and measure the frequency of the azole resistant strains in the environment as that would guide the curb of resistant species.
