EXTRACTION AND CHARACTERIZATION OF ALPHA AMYLASE BYASPERGILLUS NIGER USING YAM PEELS AS SUBSTRATE
ABSTRACT/DESCRIPTION
Extraction and characterization of Alpha amylase by Aspergillus niger using yam peels as substrate holds tremendous fungal potentiality in high biomass yield of alpha-amylase. Isolation of alpha amylase from Aspergillus niger using yam peels as substrate in a solid state fermentation medium was carried out for days. The crude enzyme was extracted from fermented media by adding 100ml of 100mm tris buffer pH 6.2 agitating the flask at 180rpm for one hour, the mixture was filtered and centrifuged at 8000rmp at 40oC for five munites and the supernatant collected as the crude enzyme. The enzyme assay was read at 0Do540nm and enzyme activity found to be 0.08u/min. the concentration of the crude enzyme was 0.187mg/ml. this study shown that renewable energy sources such as yam peels can be good source of substrate for Aspergillus niger in production of Alpha amylase.
